this is clear now. Considering your spatial resolution (0.25 deg), a river reach within a grid cell should have, on average, 25-30km (let's use 25,000m). Let's say it's a large river with 1000-meter width, and a few meters depth, say 2 meters. 25,000 x 1000 x 2 = 50 million m3. Your 15 million m3 seems reasonable.
