Validate amino acids used; fail with informative message if we can't handle them

Right now, tensorflow fails on looking up an unknown amino acid character, something like

```
= -1 is not in [0, 26)
         [[{{node GatherV2}}]]
```

Instead, we should print a nicer error to let people know what the problem is
