FIRED - Fura-2 Imaging based evaluation of dorsal Root ganglia Excitation Dynamics
This repository contains MATLAB code for a graphical user interface (GUI) that evaluates calcium imaging traces over time on a single cell level. The traces were acquired and normalized using ImageJ. All traces underwent normalization against background fluorescence signals, which were obtained by averaging 5 regions of interest (ROIs) distributed across 4 quadrants of each experimental image sequence.
- Single cell trace labeling and automatic analysis over time
- Detailed analysis for each application period
- Automatic readout of peak-normalized intensity, peak ratios, and peak duration (in development)
- Exclusion of non-viable cells from analysis
- Baseline correction for photobleaching-induced drift (optional)
- Single cell traces - Individual cell responses over time
- Multi-cell trace analysis - Combined traces with mean values and standard deviation displayed as shaded areas
- Heatmap visualization - All traces from a single experiment displayed in heatmap format
- Network plots - Dynamic visualization of response type changes based on toxin application
