Repository for the article: “High-throughput methods enabling random duplications, deletions or nucleotide-constrained mutagenesis of entire DNA motifs” by
Marcus Wäneskog, Knud Nor Nielsen, Marcus Deichmann, Samuel Alan Bradley, Michael Krogh Jensen, Emil Damgaard Jensen.
This repository contains the analysis of the PacBio Revio deep sequencing used to validate and to characterize all mutations generated by the Random Duplication, Deletion and Insertion Mutagenesis (RADDIM) technique. The analysis focuses on quantifying the types and the frequencies of mutations introduced into a control plasmid by RADDIM.
This repository also contains the Python scripts used to calculate the theoretical mutational landscapes that can be achieved using the Nucleotide-Specific Mutagenesis (NSM) technique.
